Issue
After setting a group to be challenged or exempted from challenge in the via Control Panel > RSA Authentication Agent, the update to the challenge settings update is not working.
Cause
The RSA Pipe Service needs to be restarted.  Running just an iisreset is not sufficient.
Resolution
Restart both the RSA Pipe Service and RSA Pipe Service32 through services.msc interface:

Workaround
As an alternative to restarting the two pipe services, you can reboot the Windows server as whole
